光热剂Cy7-TCF对人胰腺癌细胞氧化应激及凋亡的影响

Abstract:目的 探讨光热剂Cy7-TCF对人胰腺癌细胞氧化应激及凋亡的影响。方法 将人胰腺癌细胞系Mia PaCa-2细胞分为对照组、Cy7-TCF组(仅加Cy7-TCF)、Cy7-TCF+激光组(Cy7-TCF处理后再行808 nm激光照射)。采用CCK-8法评估不同浓度Cy7-TCF在有无激光照射下的细胞活性,通过荧光显微镜观察细胞对Cy7-TCF的摄取能力,利用共聚焦显微镜结合溶酶体探针分析Cy7-TCF的亚细胞定位,采用DCFH-DA荧光探针检测细胞内ROS水平,通过流式细胞术检测细胞凋亡率。结果 CCK-8法检测结果显示,在激光照射下,随着Cy7-TCF浓度升高,细胞存活率显著降低,呈现浓度依赖性(P<0.05);荧光显微镜观察结果显示,Cy7-TCF可被Mia PaCa-2细胞有效摄取;共聚焦显微镜观察结果显示,Cy7-TCF主要定位于溶酶体内;DCFH-DA探针检测结果显示,808 nm激光照射下,Cy7-TCF显著诱导了Mia PaCa-2细胞内ROS水平升高(q=19.07、16.84,P<0.05);流式细胞术检测显示,Cy7-TCF+激光组的凋亡率显著高于对照组及Cy7-TCF组(q=28.78、24.89,P<0.05)。结论Cy7-TCF可被胰腺癌Mia PaCa-2细胞有效摄取并定位于溶酶体内,通过光热效应诱导细胞中ROS水平升高,降低细胞存活率并促进细胞凋亡。
