Low biofouling strategy for simultaneous determination of two proteins to one tumor in human serum

摘要:Simultaneous testing of multiple tumor proteins to one tumor is essential to increase the accuracy of early cancer screening to avoid false positives. Herein, an antifouling platform based on poly guanine (poly-G) was adopted for accurate, simultaneous assay of two lung cancer-associated proteins, neuron-specific enolase (NSE) and carcinoembryonic antigen (CEA) in real samples. In this fluorescence resonance energy transfer (FRET)based strategy, MoS 2 -Cys nanospheres were used as a single energy acceptor, and two carbon dots (b -CDs, y -CDs) of different color, which was excited at one wavelength, were used as energy donors. Two types of tumor- proteins can then be simultaneously distinguished and quantitated, obtaining the detection limits (3 sigma /k ) of 0.097 ng mL -1 and 0.089 ng mL -1 for NSE and CEA, respectively. Results cor well with data from electrochemiluminescence (ECL) immunoassays in hospitals, demonstrating a clinically relevant performance for NSE and CEA assay in the unprocessed and undiluted human serum, proving to be a promising clinical diagnostic strategy for lung cancer.
